Reciprocal crosses between Lolium multiflorum plants which were either tolerant (cv. Axis) or susceptible to crown rust were performed and the segregation of the tolerant phenotype in the Fl and G2 progeny was analysed (Adams et al., submitted). In all the crosses where the tolerant genotype Axis-2 was involved, a maternal type of tolerance transmission was observed. One of these crosses (Axis-2 x B-1) was screened to search for markers linked to the tolerance at organelle DNA level. First of all an AFLP based protocol on organelle DNA was developed. The developed technique was used to study organelle DNA of the cross between Axis-2 and B-1. Several polymorphic bands were revealed. The origin of the bands needs further investigation.
