Drought stress and its effects on plant productivity are receiving more interest. In this study the quantitative expression pattern of genes inducing stress adapatation to water deficit is investigated in Fragaria sp. Two Fragaria species together with one tolerant and one sensitive cultivar were selected and exposed to drought stress for gene expression analysis. The selected candidate genes are commonly regulated by water deficit and encode the key enzymes of different pathways like osmotic adjustement, ascorbic acid metabolism and the genes encoding for some anti-oxidant enzymes? Also three involved transcription factors in plant response to drought stress were examined. Genomic/cDNA sequences of Fragaria corresponding to these putative genes were cloned and sequenced. The cloned sequences shared a high homology with functional genes in Prunus and/or Arabidopsis. Reliable reference genes were identified by investigation of variability in transcript accumulation of 11 cloned genomic sequences corresponding to frequently used reference genes. the geNorm software was applied to determine the most stable reference genes for RT-qPCR normalization in Fragaria during drought stress. Finally the expression pattern of genes of interest was analyzed using RT-qPCR under drought stress.
