The use of rhizogenic Agrobacteria as an alternative breeding technique in ornamentals

    Rhizogenic Agrobacteria are commonly used in plant transformation protocols because of their natural DNA transfer ability. Rhizogenesis inducing strains modify the host plant nuclear genome by inserting oncogenes (rol genes). Expression of these rol genes results in excessive formation of adventitious roots which are referred to as hairy roots. Plants regenerated from hairy roots typically display dwarfism, extensive lateral branching and reduced apical dominance. A more compact growth is an important aspect in the breeding process of many ornamentals. Obtaining genetic and sustainable compact growth by traditional breeding is both a stretched-out and complex process. In a contemporary production environment, compact growth is achieved by regular application of various chemical growth inhibitors. Therefore, introduction of rol genes through co-cultivation is a promising technique for attaining compactness and reducing the use of chemical growth inhibitors. Our project aims to introduce the rol genes into new genotypes of 2 ornamental species, Osteospermum sp. and Gloxinia sp. Since symptoms of the hairy root phenotype vary depending on the strain used for transformation, different strains were collected and their growth pattern was studied. In this way we could select very diverse strains to obtain a broad range of phenotypic variations in both species. Here we report on the bacterial strains and explants used and relate this to the success rate of rol gene introduction.
